摘 要:本研究通过一系列单因素试验(培养基组成、红曲霉培养时间即菌龄、渗透压稳定剂种类和浓度、酶系统的组成以及混合酶的浓度、酶解时间等)对红曲霉原生质体的形成与再生条件进行了研究。结果表明:以豆芽汁培养液为菌丝培养基,采用60h菌龄的菌丝体,以0.6mol/L MgSO4作为渗透压稳定剂,用1.5%混合酶(蜗牛酶:纤维素酶=7:3) 在30℃下处理2h为最佳的红曲霉原生质体制备条件,此条件下,原生质体形成数可以达到1.61×107个/ml,再生率为8.49%。

Abstract:A series of experiments (including culture medium, the culture time of mycelium, kinds of osmotic stabilizer and its concentration, the proper enzymes and their proportion, enzyme hydrolyzing time) were conducted to study the optimun condition of Monascus rubber protoplast. Results indicated that suitable culture medium was PDA, the culture time of mycelium was 60-day-age, the proper enzymes were snail enzyme and cellulose enzyme, the proper concentration was 1.5% (snail enzyme: cellulose enzyme=7:3), and the osmotic stabilizer was the 0.6mol/L MgSO4. Under the above conditions, the protoplasts formation was 1.61×107/ml, and the regeneration rate was 8.49%.
