摘 要:通过单因素实验和正交实验,研究了枯草芽孢杆菌(Bacillus subtilis)的液体发酵培养基和工艺条件,并对原始菌种发酵培养基的碳源、氮源等的选择及配比进行调整,以确定其各项要素的合理配比。确定最佳的培养基为:蔗糖3% ,蛋白胨1% ,酵母膏0.5% ,KH2PO4 0.45%,(NH4)2SO4 0.25% ,CaCO3 0.2% ;并对培养基单一发酵条件的改变,通过培养结果得出该条件下的最佳值,应用于具体实验操作中去,最终获得温度、pH值、装量和接种量四项发酵因素下的最佳组合,并对该组合进行验证,证明该最佳组合是否能真正达到提高菌种发酵培养的目的。实验证明最适发酵条件为:温度为30%,pH值为7.0,装量为20ml/250ml锥形瓶,接种量为5%,转速200r/min条件下。

Abstract : The single factor and orthogonal experiment. of Bacillus subtilis (Bacillus subtilis) and the liquid fermentation medium conditions. primal bacterium medium carbon, nitrogen ratio and the choice of adjustment to determine the elements of a reasonable ratio. Determine the best medium : 3% sucrose, 1% peptone, yeast extract 0.5% KH2PO4 0.45% (NH4) 2SO4 0.25%, 0.2% CaCO3; also a single medium fermentation conditions change, the results of the conditions through the cultivation of the best values application specific experimental operation, and eventually the temperature, pH value, inoculum load and the best combination of four factors fermentation, and this combination of certification, proved the best mix of bacterium can truly improve training purposes. Experimental evidence for the optimum conditions : temperature 30%, pH 7.0, 20ml/250ml installed capacity of tapered bottles inoculation of 5% speed agitation conditions.
