摘要:为了提高枯草芽孢杆菌黑色变种ATCC 9372的芽孢形成率,在最佳培养基确定的基础上,研究培养条件,包括初始pH、培养温度、接种量和溶氧水平,对芽孢形成率的影响。试验结果表明,最佳的培养条件为培养基(葡萄糖5g,蛋白胨5g,NaCl5g,MnSO4·H2O0.04g),于pH7.0,接种量5%,装瓶量为25ml/250 ml锥形瓶,30C摇床培养24小时,此时,芽孢数量最多可达到3.4×107cfu/ml,芽孢形成率达到94.4%。

Culture Conditions Affecting Spore Production of Bacillus subtilis var. niger
Abstract: In order to enhance spore production of Bacillus subtilis var. niger ATCC 9372, on the basis of optimal medium, culture conditions, including initial pH、temperature and dissolved oxygen levels on its spore production were examined. Results suggested that the optimal medium composition for spore production was as follows lactose 5g, peptone 5g, NaC1 5g, MnS04·H20 0.4 g, H20 1000 ml, optimal initial pH was 7. 0 and appropriate medium volume was 25 ml in 250 ml flask. The number of spores could reach 3.4 x 107 cfu/m1, and the highest spore formation was 94.4% at 30℃, 150 r/min for 24 h.
