提要 寻找与高血压以及血压调节有关的基因,已成为当前心血管领域的研究焦点之一,一些属于单基因遗传病的继发性高血压的致病基因已被识别。原发性高血压是一种多基因遗传病,其发病系遗传与环境因素共同作用的结果,这为高血压易感基因研究带来很大困难。迄今大多用候选基因法进行高血压相关基因研究。在人和动物模型的研究中,有关高血压候选基因多态性的报道已有四十多种,但各家结果不一致。近年来用微卫星标记进行全基因组扫描和连锁分析,它已成功地用于多基因病相关基因的定位克隆研究。应用该技术对原发性高血压易感基因的定位研究正在进行中。
Gene studies in hypertension:State-of-art
Abstract The search for genes influencing blood pressure or hypertension has become one of the focal point of cardiovascular research. A few disease genes causing secondly hypertension,as a result of a single gene defect, have been identified. However,essential hypertension is a polygenic disease and both genetic and environmental factors play important roles in its development. This fact makes the identification of its susceptibility genes more difficult. So far most studies to reveal the genes related to essential hypertension have employed a candidate gene approach.Using this strategy,the associations between polymorphisms of over forty genes with essential hypertension have been reported in human or in animal models.However,the results are not consistent .Recently,a genome-wide scan and linkage analysis using microsatellite markers becomes a useful approach in the positional cloning of genes in polygenic diseases.Its application for mapping susceptibility genes of hypertension is ongoing.
