摘要:本文详细论述了蟾皮的甲醇提取工艺,所用到的试剂、仪器,采用的方法和操作要点。通过文献考察,知干蟾皮中所含的蟾毒灵、华蟾酥毒基、蟾毒配基等成分均为极性相对比较小的内酯类化合物。通过甲醇加热回流提取与甲醇超声提取对比实验,确定出甲醇超声提取方法。通过对实验结果分析,优化实验方案。采用HPLC法以华蟾酥毒基为指标对提取液进行检测,流动相对比了药典方法0.5%磷酸二氢钾溶液—乙腈流动相系统与文献方法乙腈—水流动相系统。最终对药典方法进行比例的调整,即能达到较好的分离,分离度≥1.5,理论塔板数按华蟾酥毒基计不低于2000 。

Abstract:. This article discusses the process of the methanol extraction of toad skin in detail, the use of reagents and equipments, the methodology and operation of the main points., It is said in literature that Dry skin contains Xenopus Bufalin, cinobufotalin ,China Chansu drug-based, which are lactone compounds with small polarity .. Comparing the heating circumfluence extraction with the ultrasonic extraction of methanol , the latter was chioced in the experiment. Through different tests and the analysis of the experimental results, an optimized experiment program was determined. Cinobufotalin as the study goals from extract by HPLC .The mobile phase was determind by contrast the ChP method ( solution of 0.5% potassium dihydrogen phosphate - acetonitrile mobile phase system) with the literature method ( acetonitrile - water mobile phase systems). A better result can be acquired by pharmacopoeia method with adjusting the ratio of mobile phase. The resolution is over 1.5, and theoretical plate number in accordance with Cinobufotalin is not less than 2000.
