摘 要:酶的催化混杂性,是当今催化领域研究的热点,它是指同一个催化核心具有催化两个或者两个以上不同类型的化学反应的能力。考察酶的混乱性催化,为进一步研究酶的结构与功能的关系和开发酶的新功能提供了新的思路, 同时为蛋白质科学研究开辟了新的空间。本次实验通过筛选数种酶对 knoevenagel 反应的催化能力,第一次发现猪胰脂肪酶对这个反应具有很好的催化作用。并且,我们发现脂肪酶在催化这个反应的同时,还可以完成它原有的酯交换的能力。通过对这两种反应的动力学研究,我们发现了脂肪酶催化 knoevenagel 反应的能力远远强于其固有的催化酯交换的能力。本次实验不仅扩大了酶的催化领域,而且对酶的进化具有一定的意义。

Abstract:Nowadays,the hybridity of catalysis of enzyme is the focus areas of catalytic current. It is a capability which can catalyst two or more different types of chemical reactions by the same catalytic core. The expedition of the hybridity of catalysis of enzyme,provides a new way of thinking for father studying of the relationship between structure and function of the enzyme and developing the new functional of enzymes. By screening several enzymes which have the ability of catalytic knoevenagel reaction,we find the PLL has a very good catalysis to this reaction at the first time. In the reaction,we find that Lipase can not only catalytic the reaction,but also execute the original reaction of transesterification. Through the study of Dynamics of these two reactions,we find that the capability of the enzyme catalytic the reaction is faster than the reaction of transesterification. The experiment not only expanded the field of enzyme catalysis,but also have a certain significance to the evolution of enzyme.
