人造血干细胞扩增的研究进展

progress on Study of Techniques for Expansion of Human Hematopoietic Stem Cells
 
 造血干细胞(llematopoietic stem cells,HSC)移植可能使遗传血液病、免疫缺陷病及恶性肿瘤等完全治愈,由于HSC移植存在以下不足而受限制:⑴为获得足够量移植用HSC,必须进行大规模的骨髓抽吸或餐周血分离;⑵获得的HSC量有限;⑶HSC输注后产生的成熟细胞的生物动力欠佳,移植后1-3周内并无直接治疗疗效。移植前HSC培养扩增,可解决这些难题。当前主要有两种:⑴细胞因子支持下筛选CD34+细胞的体外扩增;⑵基质支持下的灌注培养。下面就HSC培养的发展、HSC扩增方法以及展望等作一综述。

1 早期HSC培养及启示
  1半固体培养体系 1966年,Bradley等介绍了一种新的半固体培养方法(即集落形成法)用以培养骨髓。此法利用胶体凝胶作支托,没有细胞微环境,只能维持造血祖细胞在1-2周内形成细胞集落,不能支持HSC的生长或分化。即使没法加入营养物、生长因子等,培养时间也只能延长到2-4周。由半固体培养体系的期限性可推断,早期原始HSC不能在这样的培养体系中存活、增殖。
